Microestrutura e ultraestrutura do vitelário do inseto cochonilha, Dactylopius coccus (Hemiptera: Dactylopiidae)

Resumo

O inseto cochonilha, Dactylopius coccus, é utilizado para a obtenção de ácido carmínico. Para contribuir com a compreensão da morfofisiologia do ovário dessa importante espécie, descrevemos a microestrutura e a ultraestrutura do vitelário durante a vitelogênese. Seus ovaríolos telotróficos são constituídos pelo germário, vitelário e pedicelo. Cada vitelário contém um único folículo ovariano, composto por células foliculares e oócito, e o folículo é revestido externamente por uma túnica própria. As células foliculares eram colunares a cuboides, mononucleadas e continham abundantes ribossomos e mitocôndrias, embora com pouco retículo endoplasmático rugoso. O oócito continha um grande número de mitocôndrias, ribossomos e microtúbulos, mas pouco retículo endoplasmático rugoso e complexos de Golgi. O citoplasma cortical do oócito exibia lamelas anulares, formadas a partir de um corpo fibrogranular. A gema era composta principalmente de grânulos proteicos e glóbulos lipídicos, sendo as proteínas as primeiras a serem incorporadas endocitadamente no oócito. Dentro de suas membranas endocíticas, o material proteico se compacta até a completa formação dos grânulos de gema. Corpos lamelares foram observados próximos a esses grânulos ou suas membranas, sugerindo seu papel na reciclagem dessas membranas. Os oócitos continham bactérias simbióticas, que são transferidas do germário para o oócito através do cordão trófico, pelo menos desde o início da vitelogênese. O estudo de algumas das características microestruturais e ultraestruturais do vitelário, e até mesmo do germário, é importante para o estabelecimento de relações filogenéticas entre os diferentes grupos de Coccomorpha.
