Microstructure and ultrastructure of the vitellarium of the cochineal insect, Dactylopius coccus (Hemiptera: Dactylopiidae)

The cochineal insect Dactylopius coccus is used to obtain carminic acid. In order to contribute to the knowledge of the morphophysiology of the ovary of this important species, the microstructure and ultrastructure of the vitellarium during vitellogenesis were described. Dactylopius
coccus ovarioles, of telotrophic type, are formed by the germarium, the vitellarium and the pedicel. Each vitellarium contains a single ovarian follicle, consisting of the follicular cells and the oocyte, being the follicle externally covered with a tunica propria. The follicular cells were columnar to cubic, mononucleated and with a large number
of ribosomes and mitochondria, although scarce rough endoplasmic reticulum. The oocyte showed a great amount of mitochondria, ribosomes, microtubules, but scarce rough endoplasmic reticulum and Golgi complex. The cortical cytoplasm of the oocyte presented annular lamellae, formed inside a fibrogranular body. The yolk was made mainly
of protein granules and lipid globules, with proteins being the first to be endocytically incorporated into the oocyte; within their endocytic membranes the proteinaceous material is compacted until the complete formation of yolk granules. Near the granules or their membranes,
lamellar bodies were observed, suggesting that these participate in recycling the membranes. The oocytes had symbiont bacteria, which are transferred from the germarium to the oocyte through the trophic
cord, at least since the onset of vitellogenesis.
