Protected natural areas have a key role in the fight against the loss of diversity that we are currently experiencing. This work is the first study of medium and large mammals’ assemblage (MLM) for the future protected area “Islas y Canales Verdes del Río Uruguay” (IyCVRU), Entre Ríos, Argentina. In this study, we worked on the three largest islands through monitoring with camera traps. A systematic sampling (SS) was carried out by placing three cameras on each island, separated by one kilometer from each other (nine in total), and a participatory sampling (PS) with eight cameras placed by local residents, in sites of their choice. The cameras monitored three vegetation units: Coastal Albardón Forests (CAF), Internal Albardón Forests (IAE) and Esteros Shrublands (ES). The objective of this study was to evaluate the MLM in terms of richness, diversity, relative abundance, use of environments and islands, compare between methodologies and evaluate temporal patterns. It was possible to detect nine mammal’s species, three exotic and six natives. Among the three islands monitored, the island with the least anthropic intervention was different from the other two, with greater richness and number of records. Although the assemblages between environments do not show variation, the highest richness was in BAI. Eight species were detected in SS and nine in PS. It was possible to evaluate the temporal patterns of eight species. These are the first contributions to the management and conservation for MLM for the future protected area IyCVRU.
